人脐带静脉灌注液间充质干细胞的分离和鉴

【摘要】本研究旨在探讨人脐带静脉灌注液中是否存在间充质干
细胞及其分离和扩增方法。从正常产妇分娩后的脐带静脉灌注液分离
间充质干细胞,观察细胞形态并用流式细胞仪分析其细胞表型和细胞
周期,在体外分别诱导其成骨、成脂肪和成软骨细胞分化。结果表明:
脐带静脉灌注液来源的间充质干细胞为成纤维细胞样,高表达 CD29、
HLA ABC、 CD166、 CD105、 CD73、 CD44; 不表达造血和内皮标志 CD34、 (
CD45、CD144 和 CD14) 它们可向骨、脂肪和软骨细胞分化,符合间充 。
质干细胞的基本功能特征。结论:人脐带静脉灌注液存在间充质干细
胞,能够在体外培养和扩增,可作为今后细胞治疗和组织工程的种子
细胞。

Isolation and Identification of Mesenchymal Stem Cells
from Perfusion of Human Umbilical Cord Vein
Abstract This study was aimed to investigate whether

mesenchymal stem cells (MSCs) existed in human umbilical cord
vein and to establish the methods of isolation and expansion in
vitro. The MSCs direved for perfusion of umbilical cord vein
(UVMSCs) were collected after parturition of Healthy pregnant
women. The morphology of MSCs and their differentiation
potential into osteoblast, adipocyte and chondrocyte were
observed the phenotype and cell cycle of MSCs were determined
by using flow cytometry. The result showed that the mesenchymal
stem cells separated from umbilical cord vein gave rise to a
population of adherent cells with a typical fibroblast like
morphology. Similarly to bone marrow derived MSCs, they highly
expressed CD29, HLA ABC, CD166, CD105, CD73 and CD44, and were
negative for any hematopoietic and endothelial markers (CD45,
CD34, CD14 and CD144). Functionally, they could differentiate
into the osteoblast, adipocyte and chondrocyte. It is concluded
that MSCs exist in the human umbilical cord vein perfusion. Their
read amplification in vitro contribute to clinical applications
for cell therapy and tissue engineering.

间充质干细胞(mensenchymal stem cells, MSC)最早自骨髓中
分离,是一类具有自我更新及多向分化潜能的 中胚层源干细胞[1] 。
作为多种骨髓基质的前体细胞,MSC 能通过调节造血微环境以促进造
血干细胞增殖、分化,它亦能调节淋巴细胞免疫功能,从而减轻移植
排斥反应等生物学效应[2] MSC 的发现为针对性解决造血干细胞移 。
植,尤其是脐血移植中的干细胞数量相对不足及减轻移植物抗宿主病
提供了一个有效手段[3,4] 然而,临床上骨髓取材困难,供体有限, 。
随年龄的增加其增殖和分化能力下降,且有病毒污染的可能[5] 。
因此,寻找 MSC 的新来源具有重要意义。本研究从易于获取、且组织
来源较为原始的脐带静脉灌注液中寻找 MSC(UVMSC) 并对其进行生 ,
物学特性的鉴定。
材料和方法
材料与试剂
脐带来自北京市中西医结合医院正常分娩产妇, 均经父母授权同
意。Ⅳ 胶原酶为 Sigma 公司产品,a DMEM 为 Hyclone 公司产品,
胎牛血清为 FBS,Stem Cell 产品,L 谷氨酰胺为 Amresco 公司产品,
EGF 为 Peprotech 公司产品,rh VEGF 为 R&D 公司产品,bFGF
为双鹭药业公司产品,胰蛋白酶为 Amresco 公司产品,诱导试剂:地
塞米松、b 甘油磷酸钠、抗坏血酸磷酸盐、3 异丁酸 1 甲基
