中草药叶下花总黄酮提取方法

【摘要】 目的对叶下花总黄酮的种类与提取方法进行初步研

究。方法采用定性检测、光谱分析、单因素测定、正交实验等，研究

黄酮种类，考察乙醇体积分数、温度、固液比、时间对提取率的影响。

结果叶下花含黄酮、黄酮醇、二氢黄酮、二氢黄酮醇等多种黄酮类化

合物；所考察的影响因素中，对总黄酮提取率影响程度大小顺序为乙

醇体积分数＞温度＞时间＞固液比。结论最佳提取条件为 A1B2C3D3

(乙醇体积分数 30%、温度 65℃，提取时间 180 min，固液比 1∶80)，

在此提取条件下，提取量高达 5.233%。

Abstract ： ObjectiveTo optimize the extraction

conditions for the total flavonoids from Ainsliaea pertyoides

Franch and to study the categories of the total flavonoids.

MethodsThe methods of the chemical qualitative detection, the

spectral analysis, single factor determination, orthogonal test

were adopted to study the categories of the total flavonoids,

and the effect of four factors, i.e. the volume fraction of

ethanol, the temperature, the ratio of solid to liquid, the

extracting time, on the rate of the total flavonoids, were also

investigated at the same time. Results1)The total flavonoids

consisted of flavones, flavonols, flavanones, flavanonols in A.

pertyoides. 2)The order of factors to affect the flavonoid

extraction was volume fraction of

ethanol>temperature>extracting time>ratio of solid to

liquid. ConclusionThe optimum extraction conditions are

A1B2C3D3. The highest extraction rate of the total flavonoids

reached 5.233% under the optimal conditions.

叶下花 Ainsliaea pertyoides Franch. var.

albo-tomentosa Beauverd 也称白背兔耳风、白背叶下花、追风箭等，

菊科(Compositae )帚菊木族(Mutisieae Cass)兔耳风属(Ainsliaea

DC)草本植物 1］ 是 ［ ， 《本草纲目》 等多部药典收录的天然药用植物 1］ ［。

叶下花作为骨科良药分布广泛、资源丰富、临床应用和民间用药历史

十分悠久，并能用于治疗强直性脊柱炎，填补了中药治疗在此领域的

空白［2］ ，意义重大。本研究小组发现［3］ ，叶下花中黄酮类化合物

具有明显的抗氧化活性和羟基自由基清除能力，但不清楚叶下花作为

骨科良药的活性成分是否与此有关。已有很多文献指出，高等植物中

类黄酮有多种药理作用，是一类十分重要的天然有机化合物，其中很

多接骨效果很好的药用植物，其活性成分就是黄酮类化合物［4～8］ ，

所以本工作首先对叶下花总黄酮提取方法进行研究，以期为叶下花黄

酮药理活性及叶下花中药现代化的研究奠定基础。

1 材料与仪器

1.1 中药材叶下花干燥全草于 2006-10 采自云南省楚雄市树

苴镇，洗净、风干、粉碎、过 40 目筛，备用。

1.2 仪器与试剂

Unic 7200 型分光光度计［尤尼柯(上海)仪器有限公司］ ；

Bs210s 电子天平(北京赛多利斯天平有限公司)；HWS12 型电热恒温浴

锅(上海一恒科技有限公司)；RE-2000 型旋转蒸发仪(上海亚荣生化仪

器厂)；RO-DI-50-RE 纯水系统(厦门锐思捷公司)。

芦丁(rutin)标准品，成都欧康植化科技有限公司产品。FeCl3、

NaNO2、Al(NO3)3、NaOH、盐酸、Mg 粉、无水乙醇等均为分析纯，实

验用水为去离子水。
