东亚钳蝎提取物对癫痫小鼠大脑皮层 NMDA
受体和 GABAA 受体的调节作用

摘要: 目的研究 BmKE 对抗戊四氮致癫痫作用及其对致痫小鼠
大脑皮层 NMDAR 和 GABAAR 的调节作用。方法观察戊四氮(PTZ)致癫
痫模型小鼠,丙戊酸钠(VAP,阳性对照)治疗组和 BmKE 低(L) 、中
(M) 、高(H)剂量治疗组小鼠癫痫发作的潜伏期,应用同位素 3H 分
别标记 MK-801 和 Mucimol,检测小鼠大脑皮层 NMDAR 和 GABAAR 的结
合活性。结果 BmKE(M)能显著延长 PTZ 致癫痫小鼠的发作潜伏期(P
<0.05),BmKE(L)作用很小,BmKE(H)治疗组癫痫小鼠发作潜伏期
的延长值比 BmKE(M)治疗组低。PTZ 显著升高致痫小鼠大脑皮层的
NMDAR 结合活性(P<0.05) BmKE(L)治疗组小鼠大脑皮层 NMDAR 结 ,
合活性降低,但是无显著性差异,BmKE(M)和 BmKE(H)均极显著降
低癫痫小鼠大脑皮层 NMDAR 结合活性(P<0.001) BmKE(L)和 BmKE 。
(H)显著提高 GABAAR 结合活性(P<0.05) BmKE(M)极显著提高 ,
GABAAR 结合活性(P<0.001) 结论适量的 BmKE 能够有效治疗戊四氮 。
引起的癫痫,显著延长癫痫小鼠的发作潜伏期,BmKE 抗癫痫的作用机
理,可能与其抑制 NMDAR 结合活性,激活 GABAAR 的结合活性有关。

The Modulation of Buthus martensic Karch Extract(BmKE)
on Cerebral Cortex NMDA Receptor and GABAA Receptor in Epileptic
Mice
Abstract:ObjectiveTo study the antagonism of BmKE on
epilepsy of PTZinduced epileptic mice and the modulation on NMDA
receptor(NMDAR) and GABAA receptor (GABAAR) binding activities
in cerebral cortex. MethodsThe convulsion latent periods of
PTZinduced epileptic mice, vaproate sodium VAP,

groups were observed. NMDAR and GABAAR binding activities of
cerebral cortex were detected by isotope 3H-tagged MK-801 and
mulimol respectively. ResultsBmKE(M) significantly prolonged
laten periods of epileptic mice(P<0.05), BmKE(L) had a few
effect on latent periods, the prolonged value of latent periods
treated by BmKE(H) was shorter than that treated by BmKE(M). PTZ
significantly increased NMDAR binding activities in cerebral
cortex of mice (P<0.05). Both BmKE(M) and BmKE(H) excessively
decreased those of PTZ-induced epileptic mice(p<0.001),but
those of BmKE(L) treated group had no significant difference
with those of PTZinduced epilepsic mice. GABAAR binding

activities were reduced by PTZ. (P<0.005). BmKE(L) and BmKE(H)
significantly increased those of PTZinduced epileptic mice(P
<0.05), and BmKE(M) excessively increased those of PTZinduced
epileptic mice(P<0.001).ConclusionSuitable BmKE can prolong
latent periods of epilepstic mice induced by PTZ, and has an
antiepileptic effect. The effects of BmKE may have relationship
between the inhibition on NMDAR binding avtivities and
excitation on GABAAR binding avtivities in cerebral cortex of
epileptic mice.

东亚钳蝎 Buthus martensic Karch 是我国传统药材,中医临
床用干燥全虫治疗惊风抽搐、风湿痹痛等症[1] 提取分离有效部位, 。
进而纯化有效成分,对其进行药理活性的检测,研究其作用机理将进
一步提高其临床应用价值。在实验性癫痫中,GABAAR 的 Cl-通道阻断
剂戊四氮(pentylenetetrazol,PTZ)被广泛应用于诱导产生惊厥、建
立癫痫动物模型。早期的研究发现,PTZ 不仅具有抑制 GABAA 受体的
效应[2,3] ,连续注射还可引起 NMDA 受体激活,导致 Ca2+向细胞内
流动[4] 。东亚钳蝎在中医临床上常用于同其他药物配伍,控制癫痫
的发作,前期研究发现,BmKE 能够延长癫痫发作的潜伏期,显著升高
