RIDASCREEN Sulfonamide ist ein kompetitiver Enzymimmunoassay zur quantitativen Bestimmung von Sulfonamiden in Ei,Fleisch(Huhn und Schwein),Fisch,Shrimps,Honig und Milch.2.Allgemeines Sulfonamide werden in der Klber-und Schweinemast hufig als Futtermittelzu-satzstoffe eingesetzt.Zusammen mit Antifolaten(Dihydrofolat-Reduktase-Hemmer)wie z.B.Trimethoprim und Pyrimethamin werden sie in der Veterinr-medizin auch hufig zur Behandlung von Darminfektionen,Mastiden,Lungen-entzndungen und anderen systemischen Infektionen verwendet.Die Kontamination von Lebensmitteln tierischen Ursprungs,wie Milch und Fleisch,mit Sulfonamiden ist deshalb mglich und wurde auch hufig(vor allem im Ausland)nachgewiesen.Laut EU-Recht gilt die Hchstmenge fr alle Stoffe der Sulfonamidgruppe von 100 ppb in Muskel,Fett,Leber und Niere sowie von 100 ppb in Milch.RIDASCREEN Sulfonamide 09-07-16 5 3.Testprinzip Grundlage ist die Antigen-Antikrper-Reaktion.Die Vertiefungen der Mikrotiter-streifen sind mit Fnger-Antikrpern gegen anti-Sulfonamid-Antikrper be-schichtet.Zugegeben werden Standards bzw.Probelsung,enzymmarkiertes Sulfonamid(Enzymkonjugat)und anti-Sulfonamid-Antikrper.Freie Sulfonamide und enzymmarkiertes Sulfonamid konkurrieren um die Sulfonamid-Antikrper-bindungsstellen(kompetitiver Enzymimmunoassay).Gleichzeitig werden auch die anti-Sulfonamid-Antikrper von den immobilisierten Fnger-Antikrpern gebun-den.Nicht gebundenes,enzymmarkiertes Sulfonamid wird anschlieend in einem Waschschritt wieder entfernt.Der Nachweis erfolgt durch die Zugabe der Substrat-/Chromogenlsung.Gebundenes Enzymkonjugat wandelt das Chromo-gen in ein blaues Endprodukt um.Die Zugabe des Stopp-Reagenzes fhrt zu einem Farbumschlag von blau nach gelb.Die Messung erfolgt photometrisch bei 450 nm;die Extinktion der Lsung ist umgekehrt proportional zur Sulfonamid-Konzentration in der Probe.4.Packungsinhalt Mit den Reagenzien einer Packung knnen 96 Bestimmungen use RIDASCREEN Sulfonamide is a competitive enzyme immunoassay for the quantitative analysis of sulfonamides in egg,meat(chicken and pork),fish,shrimps,honey and milk.2.General Sulfonamides are widely used as feed additives,mainly for fattening of calves and pigs.Combined with inhibitors of dihydrofolate reductase such as trimethoprim,tetroxoprim,or pyrimethamine sulfonamides are also used in veterinary medicine for the treatment of intestinal infections,mastitis,pulmonitis and other(systemic)diseases.Therefore sulfonamide residues may occur in food of animal origin such as meat and milk and were found frequently abroad.According to the EU-Law a maximum residue limit for all substances of the sulfonamide-group of 100 ppb in muscle,fat,liver and kidney and of 100 ppb in milk is valid.RIDASCREEN Sulfonamide 09-07-16 13 3.Test principle The basis of the test is the antigen-antibody reaction.The microtiter wells are coated with capture antibodies directed against anti-sulfonamide antibodies.Standards or sample solution,sulfonamide enzyme conjugate and anti-sulfonamide antibodies are added.Free sulfonamides and sulfonamide enzyme conjugate compete for the sulfonamide antibody binding sites(competitive enzyme immunoassay).At the same time,the anti-sulfonamide antibodies are also bound by the immobilised capture antibodies.Any unbound enzyme conjugate is then removed in a washing step.Substrate/chromogen solution is added to the wells and incubated.Bound enzyme conjugate converts the chromogen into a blue product.The addition of the stop solution leads to a color change from blue to yellow.The measurement is made photometrically at 450 nm.The absorption is inversely proportional to the sulfonamide concentration in the sample.4.Reagents 6.Warnings and precautions for the users The standards contain sulfamethoxypyridazine,particular care should be taken.The stop solution contains 1 N sulfuric acid(R36/38,S2-26).7.Storage instructions Store the kit at 2-8 C(35-46 F).Do not freeze any of the test kit components.Return any unused microwells to their original foil bag,reseal them together with the desiccant provided and further store at 2-8 C(35-46 F).The reddish substrate/chromogen solution is light sensitive,therefore,avoid exposure to direct light.No quality guarantee is accepted after the expiration date on the kit label.Do buffer)use 50 l per well in the test 9.2.Meat(pork),egg,fish and shrimps weigh in 1 g of the homogenized sample into a centrifugal screw cap vial,add 2 ml methanol and vortex for 30 sec centrifuge:10 min/4000 g/20-25 C(68-77 F)transfer 1.5 ml of the methanolic solution into a new centrifugal vial and evaporate to dryness dissolve the residue in 0.5 ml sample dilution buffer,add 1 ml of n-hexane(or n-heptane)for degreasing and vortex for 10 sec centrifuge:10 min/4000 g/20-25 C(68-77 F)use 50 l of the lower phase per well in the test 9.3.Meat(chicken)weigh in 2 g of the homogenized sample into a centrifugal screw cap vial,add 6 ml acetonitrile/water(84:16;v:v)and shake for 10 min(with up-side-down shaker)centrifuge:10 min/3000 g/15 C(59 F)transfer 4 ml of the supernatant into a new centrifugal vial,add 2 ml 2 M NaCl,7 ml ethylacetate and shake for 10 min(with up-side-down shaker)centrifuge:10 min/3000 g/15 C(59 F)transfer the whole supernatant into a new centrifugal vial and evaporate to dryness dissolve the residue in 1 ml sample dilution buffer,vortex for 1 min,add 1 ml n-hexane(or n-heptane)and,vortex again for 2 min centrifuge:10 min/3000 g/15 C(59 F)use 50 l of the 5,(see 5.2.)vortex until complete dissolving of honey centrifuge:10 min/4000 g/20-25 C(68-77 F)Purify the whole extract with RIDA C18 column(Art.No.R2002);flow rate:1 drop/sec:rinse the column with 2 ml methanol equilibrate the column with 2 ml 50 mM sodium acetate buffer(pH 5)apply the whole sample extract rinse the column with 8 ml 50 mM sodium acetate buffer(pH 5)remove any excess fluid by pressure and flushing the cartridge with a stream of either air or nitrogen for 2 min elute sample with 1 ml methanol/sample dilution buffer(20:80;v:v)slowly dilute the eluate 1:3(1+2)with sample dilution buffer(e.g.100 l eluate+200 l sample dilution buffer)use 50 l per well in the assay 10.Test implementation 10.1.Preliminary comments Bring all reagents to room temperature(20-25 C/68-77 F)before use.A PBS-Tween buffer is needed as washing buffer,please use the washing buffer salt(pouch)contained in the kit(see 4.).Dissolve the total content of the pouch in one liter of distilled water.The ready to use washing buffer expires after approx.4-6 weeks at 2-8 C(36-46 F).Alternatively:Dissolve the contents of the pouch in 100 ml of distilled water to obtain a 10fold concentrated washing buffer.This solution expires after approx.8-12 weeks,stored at room temperature(20-25 C/68-77 F).Use one part of this concentrate and dissolve with 9 parts of distilled water to obtain the ready to use washing buffer.RIDASCREEN Sulfonamide 09-07-16 17 10.2.Test procedure Carefully follow the recommended washing procedure.Do not allow microwells to dry between working steps.1.Insert a sufficient number of wells into the microwell holder for all standards and samples to be run in duplicate.Record standard and sample positions.2.Add 50 l of each standard solution or prepared sample to separate duplicate wells.Use a new pipette tip for each standard or sample.3.Add 50 l of enzyme conjugate(red cap)solution to the bottom of each well.4.Add 50 l of antibody solution(black cap)to each well,mix gently by shaking the plate manually and incubate for 1 h at room temperature(
