Inhibitory effect of siRNA on calcineurin activity and
vascular smooth muscle cell proliferation in rats
【Abstract】 AIM: To study the changes of calcineurin
(CaN) activity and cfos expression level in the proliferation
of rat vascular smooth muscle cells (VSMCs) stimulated by
neuropeptide Y (NPY), and the inhibition of siRNA on the activity
of CaN and the proliferation of VSMCs. METHODS: VSMCs from
thoracic aorta in rats were cultured in vitro using tissuepiece
anchorage primary culture. VSMCs were treated with NPY and the
cell proliferation was observed. RNA interference (RNAi) was
used to block CaNdepended signal transduction pathway in rat
VSMCs stimulated by NPY, then the activity of CaN, the
proliferation of VSMCs and the expression level of cfos were
tested. RESULTS: NPY could increase the CaN activity, the

proliferation of VSMCs, and the expression level of cfos. RNAi
inhibited the activity of CaN, VSMCs proliferation and cfos
expression in VSMCs. CONCLUSION: CaN takes part in the
proliferation of cultured VSMCs stimulated by NPY, while RNAi
blocks the proliferation of VSMCs stimulated by NPY by
inhibiting the activity of CaN.

【摘要】目的: 观察神经肽 Y(NPY)刺激的大鼠血管平滑肌
细胞(VSMCs)增殖中钙调神经磷酸酶(CaN)活性和 cfos 表达水平的变
化,以及 RNA 干扰抑制 CaN 活性的作用. 方法: 采用组织贴块法体外
原代培养大鼠胸主动脉平滑肌细胞;NPY 刺激培养的大鼠 VSMCs 增殖;
RNA 干扰特异性抑制 CaN 的活性及其 mRNA 的表达,阻断 NPY 刺激的大
鼠 VSMCs 中 CaN 依赖的信号转导通路,观察对 CaN 活性、细胞增殖、
核内原癌基因 cfos 表达水平的变化. 结果:NPY 可增加 VSMCs 的 CaN
活性,加快细胞增殖及增加核内 cfos 表达水平. RNA 干扰抑制 CaN 活
性后,明显降低 NPY 刺激的 VSMCs 增殖及核内 cfos 表达水平. 结论:
CaN 参与了 NPY 刺激的 VSMCs 增殖, 干扰通过抑制 CaN 的活性可阻 RNA
断 NPY 诱导的 VSMCs 增殖作用.

0 引言
钙调神经磷酸酶(calcineurin, CaN)属丝氨酸苏氨酸蛋白磷
酸酶家族成员,在细胞信号传递过程由 Ca2+直接调节,具有去磷酸化
作用. 目前认为,CaN 是一种广泛分布于脑、心肌、骨骼肌、T 淋巴细
胞、血管平滑肌和心血管内皮等组织细胞中,参与多种细胞功能[1] ,
心肌肥大、血管平滑肌细胞(vascular smooth muscle cells, VSMCs)
增殖和内皮细胞功能的调节[2]. Ca2+是调节 VSMCs 功能活动的重要
信号传导递质,可介导多种神经体液因素、细胞因子等的信号传递,
刺激核内多种原癌基因表达,进而激活增殖相关基因转录,促进细胞
蛋白质合成,引起细胞增殖[3]. 但胞内 Ca2+浓度变化与核内增殖
基因活化的偶联机制尚不十分清楚. 我们采用细胞培养,RNA 干扰,
免疫组化等技术,探讨神经肽 Y(neuropeptide Y, NPY)刺激 VSMCs
后的细胞增殖,细胞内 CaN 活性和 cfos 表达水平的变化,以及 RNA
干扰抑制 CaN 活性后, NPY 刺激的 VSMCs 增殖及 CaN 活性和核内 cfos 对
表达水平的影响.
