There is no real difference between "What's your job?" and "What do you do?" thought the latter may be more common. There are also a number of other ways of asking the same question, such as "What line of work are you in?", "What job do you do?", or "What do you do for a living?" In some contexts, it may not be so polite to ask someone you don't know directly about their job unless you are already on the subject. If, for example, they are complaining about what a long and hard day they had, you might be able to then say, "Oh, what is it that you do?"
