3、dy secreted from hybridoma was studied by testing the titer and affinity. The results showed that 0.25mmol/L and 0.5mmol/L sodium butyrate and various concentrations of rhIL-6 could significantly increase the titer and affinity of anti-B. But it could not be promoted by insulin and retinoic acid. So
4、me smaller ones of the experimental cells died rapidly in the HAT medium, after which the titer and affinity of anti-B rose markedly. It is concluded the sodium butyrate and rhIL-6 could promote the titer and affinity of anti-B secreted by hybridoma, surviving clones in HAT medium could secrete more
5、 anti-B antibody.Key Words Monoclonal antibody, Blood grouping reagents, Hybridoma血型定型单克隆抗体试剂已广泛用于临床医学,法医学,考古学等工作中,与某些发达国家相比,我国血型定型试剂在效价和亲和力方面仍存在一定差距,有些企业因产品效价和亲和力达不到国家标准而致产量大幅度下降,故研究出提高抗体效价和亲和力的方法将有着深远的意义。目前国际上少数学者正致力于研究无血清培养基中杂交瘤细胞抗体分泌的影响因素1,但有关在含血清培养基中如何提高杂交瘤细胞抗体分泌报导甚少,为此我们开展了本实验。1材料1.1细胞系抗B-6-1-
6、G11由本校分子生物学研究室提供。1.2试剂牛胰岛素(40U/ml),上海生物化学试剂厂生产;RPMI1640,美国GIBCO产品;氨甲喋呤(Amiopterin, A,AR级)Fluka公司产品;丁酸钠(AR级),次黄嘌呤(Hypoxanthine, H,AR级),胸腺嘧啶核苷(Thymidine, T,AR级),均为Sigma公司产品;rhIL-6,军事医学科学院产品;维甲酸,重庆华帮制药有限公司;小牛血清,合肥奶牛厂产品。1.3仪器CO2培养箱,美国IEVCO产品;倒置显微镜,重庆光学仪器厂产品。2方法2.1抗B杂交瘤细胞的培养将冻存于液氮中的抗B杂交瘤细胞复苏后,移入盛有10%小牛血清
7、1640培养液中,置37,5% CO2培养箱中培养,待细胞长满底壁后传代,第3代细胞生长速度明显加快,此细胞即可作为实验细胞。2.2丁酸钠对抗B杂交瘤细胞抗体分泌的影响将实验细胞从培养瓶底壁吹下,洗涤2次后,用10%小牛血清1640培养液配成浓度为1105个细胞/ml的细胞悬液,移入24孔培养板,每孔1 ml,然后加入含不同浓度丁酸钠的1640培养液1 ml,观察不同浓度丁酸钠对抗体分泌的影响。2.3rhIL-6,胰岛素,维甲酸对抗B杂交瘤细胞抗体分泌的影响细胞培养及实验细胞分组同前,最后向24孔培养板中加入不同浓度rhIL-6,胰岛素,维甲酸,观察它们对抗体分泌的影响。2.4HAT筛选对抗B
8、杂交瘤细胞抗体分泌的影响HAT培养基由氨甲喋呤(A),次黄嘌呤(H),胸腺嘧啶核苷(T)组成,配制见文献2,用此培养基配制浓度1105个细胞/ml的细胞悬液,移入24孔培养板每孔2ml,置37,5% CO2培养箱中培养,待培养一定时间后逐渐更换成普通培养基,另设无HAT对照,观察HAT筛选组与无HAT组之间的差别。2.5抗B抗体效价和亲和力测定见中国生物制品规程二部。2.6数据处理采用student t检验。3结果3.1丁酸钠对抗B杂交瘤细胞抗体分泌的影响按表1分组,结果显示:0.25mmol/L和0.5mmol/L的丁酸钠组抗体效价和亲和力明显提高,但当丁酸钠浓度为1mmol/L时,杂交瘤细

G:geometric mean, n=10 ?*P0.01 compared with control(rhIL-6=0) 3.3胰岛素和维甲酸对抗B杂交瘤细胞抗体分泌的影响向抗B杂交瘤细胞中加入不同浓度胰岛素和维甲酸,结果显示胰岛素可使细胞增殖速度明显加快,抗体效价和亲和力有升高趋势,但差别无显著性;维甲酸既不能提高抗体的效价和亲和力也不能增快细胞生长。3.4HAT筛选对抗B杂交瘤细胞抗体分泌的影响按表3将

G:geometric mean, n=10 *P0.01 compared with control(HAT=0) 4讨论血库、临床医学、法医以及血液免疫学,人类遗传学方面都迫切需要高效价,高亲和力,高特异性的血型单克隆抗体定型试剂。近年来发现某些脂肪酸钠盐可提高杂交瘤细胞抗体分泌的能力,其作用机制可能是抑制组蛋白脱酰基酶引起组蛋白超酰基化,从而促进基因的表达,此实验结果显示0.25 mm
15、ol/L和0.5 mmol/L丁酸钠可明显提高抗B杂交瘤细胞分泌抗体的效价和亲和力,提示丁酸钠可促进抗体分泌,此结果与文献基本相似3。IL-6被认为是杂交瘤细胞生长因子4,我们实验显示其可明显提高抗体效价和亲和力,提示其可增加杂交瘤细胞抗体分泌,此结果也与他人研究相似。胰岛素具有加速糖和氨基酸进入细胞,促进细胞内糖代谢和蛋白质合成作用,维甲酸能影响细胞增殖和抗体分泌,我们实验未显示它们能提高抗体的效价和亲和力,这与文献结果不同5,6,可能与杂交瘤细胞间差异有关。保持杂交瘤细胞持续稳定地分泌特异性单抗具有相当重要的意义。从杂交瘤群体来看,抗体分泌不稳定的原因可能是染色体丢失,不分泌细胞过度增生,
16、我们实验发现当HAT加到细胞培养液中后,最初几天大量小体积细胞死亡,说明实验细胞有部分恢复了对HAT的敏感性,故经重筛选的细胞分泌了浓度较高的抗体。因此我们认为根据杂交瘤细胞的具体情况,适时地用培养液再处理,将有助于提高抗B抗体的分泌量。除本文研究的因素外,还有其它因素影响杂交瘤细胞抗体分泌,如谷氨酰胺通过提高细胞对ATP的利用率7,蛋白胨通过提高细胞对氨基酸的利用率8增加杂交瘤细胞抗体生成量,另外培养基的种类,培养基的pH值,培养过程中乳酸和氨的浓度也将影响杂交瘤细胞抗体分泌,但有些因素将有待进一步探讨。姚余有(安徽医科大学基础部东南公司,合肥230032)张玲玲(安徽医科大学基础部东南公司
