5、eCordintelligent chip.The Certificate of Analysis isspecific to each batch of packing material contained in theACQUITY UPLCBEH column and includes the gel batch number,analysis of unbonded particles,analysis of bonded particles,and chromatographic results and conditions.The PerformanceTest Chromatog

6、ram is specific to each individual column andcontains such information as:gel batch number,column serialnumber,USP plate count,USP tailing factor,capacity factor,and chromatographic conditions.These data should be storedfor future reference.a.Column Connectors The ACQUITY UPLCsystem utilizes tubing

7、and gold platedcompression screws which have been designed to meet stringenttolerance levels and to minimize extra column volumes.Optimized column inlet tubing(part number 430001084)issupplied with the ACQUITY UPLCsystem.The inject valve endof the tubing is clearly marked with a blue shrink tube mar

8、ker.Insert the opposite end of the tubing into the ACQUITY UPLCcolumn and tighten the compression fitting using two 5/16-inch wrenches.For information on the correct column outlet tubing,pleaserefer to the relevant detector section in the ACQUITY UPLCSystem Operators Guide(part number 71500082502).b

9、Column InstallationNote:The flow rates given in the procedure below are for a typical 2.1 mm i.d.by 50 mm length 1.7 m column.For 1.0 mm i.d columns,scale the flow rateusing the calculation shown in section III.Additional flow rate and pressureguides are provided in section VI(Additional Informatio

10、n).1.Purge the pumping system of any buffer-containing mobilephases and connect the inlet end of the column to theinjector outlet.2.Flush column with 100%organic mobile phase(methanolor acetonitrile)by setting the pump flow rate to 0.1 mL/minand increase the flow rate to 0.5 mL/min over 5 minutes.3.

11、When the mobile phase is flowing freely from the columnoutlet,stop the flow and attach the column outlet to thedetector.This prevents entry of air into the detection systemand gives more rapid baseline equilibration.4.Gradually increase the flow rate as described in step 2.5.Once a steady backpressu

12、re and baseline have beenachieved,proceed to the next section.Note:If mobile phase additives are present in low concentrations(e.g.,ion-pairingreagents),100 to 200 column volumes may be required for complete equilibration.In addition,mobile phases that contain formate(e.g.,ammonium formate,formicaci

13、d,etc.)may also require longer initial column equilibration times.c.Column Equilibration ACQUITY UPLCBEH columns are shipped in 100%acetonitrile.It is important to ensure mobile phase compatibility beforechanging to a different mobile phase system.Equilibrate thecolumn with a minimum of 10 column vo

16、with 60%methanol in water priorto introducing 60%methanol/40%buffer mobile phase).d.eCordInstallationThe eCordbutton should be attached to the side of the columnheater module.The eCordbutton is magnetized and does notrequire specific orientation.e.Initial Column Efficiency Determination 1.Perform an

17、 efficiency test on the column before using it.Waters recommends using a suitable solute mixture,as foundin the“Performance Test Chromatogram”,to analyze the columnupon receipt.2.Determine the number of theoretical plates(N)and use thisvalue for periodic comparisons.3.Repeat the test at predetermine

18、d intervals to track columnperformance over time.Slight variations may be obtainedon two different UPLC systems due to the quality of the connections,operating environment,system electronics,reagent quality,column condition and operator technique.II.COLUMN USETo ensure the continued high performance

19、 of ACQUITY UPLCBEH columns,follow these guidelines:a.Sample Preparation 1.Sample impurities often contribute to column contamination.One option to avoid this is to use Waters Oasissolid-phaseextraction cartridges/columns or Sep-Pakcartridges of theappropriate chemistry to clean up the sample before

20、 analysis.2.It is preferable to prepare the sample in the operating mobilephase or a mobile phase that is weaker(less organic modifier)than the mobile phase for the best peak shape and sensitivity.3.If the sample is not dissolved in the mobile phase,ensurethat the sample,solvent and mobile phases ar

21、e miscible inorder to avoid sample and/or buffer precipitation.4.Filter sample with 0.2 m membranes to remove particulates.If the sample is dissolved in a solvent that contains an organicmodifier(e.g.,acetonitrile,methanol,etc.)ensure that themembrane material does not dissolve in the solvent.Contac

22、tthe membrane manufacturer with solvent compatibility questions.Alternatively,centrifugation for 20 minutes at 8,000 rpm,followed by the transfer of the supernatant liquid to anappropriate vial,could be considered.b.pH Range The recommended operating pH range for ACQUITY UPLCBEH columns is 1 to 12.A

23、 listing of commonly used buffersand additives is given in Table 2.Additionally,the column lifetimewill vary depending upon the operating temperature,the typeand concentration of buffer used.For example,the use of phosphatebuffer at pH 8 or above in combination with elevated temperatureswill lead to

25、ntaining suspended particulate materials will generallyclog the outside surface of the inlet distribution frit of the column.This will result in higher operating pressure and poorer performance.Degas all solvents thoroughly before use to prevent bubble formationin the pump and detector.The use of an

26、 on-line degassing unitis also recommended.This is especially important when runninglow pressure gradients since bubble formation can occur as aresult of aqueous and organic solvent mixing during the gradient.d.Pressure ACQUITY UPLCBEH columns can tolerate pressures of up to15,000 psi(1034 bar or 10

27、3 Mpa).e.Temperature Temperatures between 20 C 55 C are recommended for operatingACQUITY UPLCBEH columns in order to enhance selectivity,lower solvent viscosity and increase mass transfer rates.Operating at the extremes of pH,temperature,and/or pressurewill result in a shortened column lifetime.III.

46、assure that thecolumns performance history is maintained in the event that thecolumn is moved from one instrument to another.At the time of manufacture,tracking and quality control informationwill be downloaded to the eCord.Storing this information onthe chip will eliminate the need for a paper Cert
