亚洲棉石系亚1号耐旱相关基因cDNA文库的构建与分析的开题报告 摘要:棉花作为全球最重要的经济作物之一,其耐旱性成为制约棉花产量和质量的重要因素之一。本研究旨在构建亚洲棉石系亚1号耐旱相关基因cDNA文库,并对文库进行分析,以挖掘与耐旱相关的基因。 首先,采用实时荧光定量PCR技术筛选出抗旱性较强的亚洲棉石系亚1号品种为研究材料,并从根、茎和叶组织中提取总RNA,通过聚合酶酶链反应(PCR)扩增并构建cDNA文库。随后,对文库进行质控,检测文库插入片段的大小、插入片段数量、平均插入片段大小等指标,确保文库质量。最后,采用Sanger测序技术对文库进行测序,获取测序数据,并对序列进行注释和比对分析,以挖掘其中与耐旱相关的基因,并进一步进行功能研究。 本研究的结果对深入了解棉花抗旱机理、开发新型抗旱材料具有重要意义。 关键词:棉花;耐旱性;cDNA文库;实时荧光定量PCR;Sanger测序 Abstract: Cotton, as one of the most important economic crops in the world, the drought resistance of which has become an important factor limiting cotton yield and quality. The aim of this study was to construct a cDNA library of drought-resistant genes related to Asian cotton strain YAZ1 and analyze the library to excavate genes related to drought resistance. Firstly, using Real-time Fluorescence Quantitative PCR technology, we selected YAZ1, a drought-resistant strain of Asian cotton, as the research material, and extracted total RNA from root, stem and leaf tissues by PCR amplification and cDNA library construction. Then, the library was subjected to quality control to ensure the quality of the library by detecting the size, number and average size of insert fragments. Finally, Sanger sequencing technology was used to sequence the library, obtain sequencing data, and annotate and align the sequences to excavate genes related to drought resistance and further study their functions. The results of this study are of great significance for in-depth understanding of the drought resistance mechanism of cotton and the development of new drought-resistant materials.
